Mito-apocynin (C11), an orally active mitochondria-targeted triphenylphosphonium (TPP)-based compound, is synthesized by conjugating the Apocynin moiety with a TPP+ cation. Mito-apocynin (C11) selectively targets mitochondria, and shows neuroprotective effect. Mito-apocynin (C11) prevents hyposmia and corrects deficits in motor function .

描述Parkinson's disease (PD) patients exhibit early non-motor symptoms that are considered “preclinical”, but are increasingly recognized for their diagnostic potential. Hyposmia (diminished ability to smell and detect odors) is an early sign of PD and other neurodegenerative diseases. Mito-apocynin-C11 is a novel mitochondrially-targeted apocynin derivative which prevents hyposmia and loss of motor function in the mouse model of PD. The presence of a highly lipophilic and delocalized cationic moiety in Mito-apocynin-C11 makes it more cell-permeable and selectively target mitochondria. LRRK2^R1441G transgenic or wild type littermates were orally gavaged with 3 mg/kg Mito-apocynin-C11 3x/wk from 3 months of age until 15 months. This treatment showed that Mito-apocynin-C11 administration markedly improved the coordinated motor function and olfactory function in LRRK2^R1441G mice. Notably, long-term treatment with Mito-apocynin-C11 prevented this hyposmia with performance remaining near the level of the wild type mice. Interestingly, the ability to remain on the spinning rod was retained in LRRK2^R1441G mice orally gavaged with Mito-apocynin-C11^[2].
