JW 55 is a potent and selective β-catenin signaling pathway inhibitor, which functions via inhibition of the PARP domain of tankyrase 1 and tankyrase 2 (TNKS1/2). JW55 decreases auto-PARsylation of TNKS1/2 in vitro with IC50s of 1.9 μM and 830 nM respectively.

描述JW55 is a TNKS1/2 inhibitor with IC50 value of 470nM for inhibition of Wnt signaling in a luciferase report assay. Inhibition of TNKS1/2 poly(ADP-ribosyl)ation activity by JW55 led to stabilization of AXIN2, a member of the β -catenin destruction complex, in a dose-dependent manner at concentration ranging in 0.1-10μM in SW480 cells post 24h, followed by increased degradation of β-catenin at 5μM for 48h. It promoted cell-cycle arrest and specifically reduced proliferation in SW480 CRC cells. JW55 inhibited canonical Wnt signaling in colon carcinoma cells that contained mutations in either the APC (adenomatous polyposis coli) locus or in an allele of β-catenin. Oral administration of JW55 at 100mg/kg reduced tamoxifen-induced polyposis formation in conditional APC mutant mice^[2].
