Col003 is a cell penetrant, selective and potent inhibitor of the interaction of Hsp47 with collagen. Col003 is the cleavage product of AK778.It competitively inhibited the interaction and caused the inhibition of collagen secretion by destabilizing the collagen triple helix.

描述Hsp47 is an endoplasmic reticulum (ER)-resident collagen-specific molecular chaperone essential for correct folding of procollagen in the ER. Thus, Hsp47 could be a potential and promising target for the management of fibrosis. Col003 is a potent inhibitor of the interaction between Hsp47 and collagen with an IC₅₀ of 1.8 μM. In vivo, col003 (100 μM) inhibited collagen secretion, triple helix formation, and accumulation in the ECM^[2].
作用机制Col003 competitively binds to the collagen-binding site on Hsp47^[2].
